Inquiring readers, One of the benefits of overseeing a long-lasting blog is the number of Jane Austen aficionados one meets via email and online. Hi Vic! I?m a 4th-great-grandson of Frank Austen, and a committed genealogist. I?ve just posted a new website dedicated to Jane Austen?s Family, which was announced to the public at last week?s JAS AGM. The address is www.janeaustensfamily.co.uk . The first content is Joan Corder?s ?Akin to Jane? ? a 1953 manuscript listing as many descendants of George and Cassandra Austen as the author could find. Joan recorded something like 320 descendants of George and Cassandra Austen, which is very good going for 1953. The biographical detail in the manuscript makes it invaluable. She could never find a publisher and the book exists only in a couple of manuscript copies, one of which is at the Jane Austen?s House Museum at Chawton. When I first began working on the site, I wasn?t sure whether it would interest anyone ? I was simply driven on by my obsession with family history ? but it?s been well received, to my delight. The Museum is pleased that they can now retire Joan Corder?s fragile original.

With the benefit of modern genealogical facilities, I?ve increased the tally from 320 to over 1200 ? all of whom are to be found on my RootsWeb site. I have to admit that I have included very little anecdotal information, it is mainly genealogy; and all details except the surname are withheld for anyone born after 1915, though I have them on my computer database.

You asked for an anecdotal example for Jane Austen?s World readers that would flesh out the details of my research. I immediately thought of James Brydges, 8th Baron Chandos of Sudeley and Elizabeth Barnard ? Cassandra Leigh?s great-grandparents. Cassandra was of course Jane Austen?s mother.

Hearing Miss Barnard was engaged to a party with a fashionable conjuror, who showed the ladies their future husbands in a glass, he by a proper application to the cunning man beforehand, and by a proper position at the time, was exhibited in the glass to Miss Barnard: clapping her hands she cried, ?Then Mr. Bridges is my destination, and such he shall be.??

This lovely anecdote was recorded in a footnote, in The Complete Peerage,under the entry for James Brydges, the 8th Lord Chandos of Sudeley. The lady in question, Elizabeth Barnard, did become his wife. Elizabeth?s father Sir Henry Barnard was a ?Turkey merchant,? a trader whose business interest was in importing from Constantinople. Her husband James Brydges was himself the Ambassador of the ?Turkey Company? (properly the Levant Company) in Constantinople from 1680 to 1686.

Elizabeth gave birth to twenty-two children. We are familiar with the mortal threat to women?s lives from childrearing ? three of Jane Austens? sisters-in-law suffered that fate. Elizabeth survived her twenty-two deliveries and lived to the age of 77. Not all of her children fared so well ? only fifteen were baptized, and of those, three sons and five daughters survived infancy. This was far from unusual ? Antonia Fraser, in her study of 17th-century woman, The Weaker Sex, stated that it was normal for only a third of children born to a large family to survive. Their eldest child, Mary Bridges, was one of the survivors. The link to Jane Austen can now be traced within a few generations. Mary married Theophilus Leigh; they were Cassandra Leigh?s paternal grandparents and the parents of Theophilus Leigh, who served as Master of Balliol College in Oxford from 1726 until his death in 1785. Theophilus Jr.?s brother Thomas Leigh married Jane Walker, and they were Cassandra Leigh?s parents. Cassandra, who married George Austen, gave birth to eight children, including Jane Austen in 1775. (And she too survived to a ripe old age.)

Rare Diseases: 5 Recent Reasons to Cheer

On Sunday morning, July 21, I faced a room of people from families with Leber congenital amaurosis (LCA), an inherited blindness caused by mutations in any of at least 18 genes. It was the final session of the Foundation for Retinal Research?s bi-annual LCA family conference, and I was there to discuss the history of gene therapy. But I zapped through that quickly, because the future is much more intriguing.

The excitement pervading the room that was palpable, following a day of scientific updates, and not only because those with young children were soon to visit Sesame World and the sights of Philadelphia.

Jennifer and Troy Stevens exemplified that hope. Two years earlier, at this conference, they?d learned that researchers had been unable to identify a mutation behind their toddler Gavin?s blindness. Now they know the name of their gene: NMNAT1. I?ll return to their story.

The star of the 2010 conference had been 10-year-old Corey Haas and an energetic young sheepdog, both cured of LCA with gene therapy. This weekend, the stars were the new programs and technologies that would allow other families to join Corey?s ? and not just those with blindness.

The rare disease community in the US collectively belies its name: at least 30 million people suffer from 7,000+ diseases, many so rare that they hover beneath the radar of big pharma. But maybe not for long, thanks to the following recent reasons to cheer:

#1: GENE THERAPY PENDING APPROVAL

On July 20, the European Medicines Agency (EMA) announced impending first approval of a gene therapy in the western world.

It?s for lipoprotein lipase deficiency (LPLD). The enzyme normally breaks down tiny triglyceride-packed globules called chylomicrons, and its absence causes episodes of very painful pancreatitis that can be fatal. LPLD is an ultra-rare disease, striking 1-2 people per million. And the only treatment is a diet so low in fat that most patients can?t stick to it.

The gene therapy, Glybera, consists of adeno-associated virus type 1 delivering an overactive variant of the LPL gene, injected into a leg muscle during a single day. But not many people have had it.

The research team, led by Daniel Gaudet, MD, PhD, a professor of medicine at the University of Montreal, with colleagues from Amsterdam Molecular Therapeutics (recently replaced by privately-held UniQure), reported a two-year follow-up of 14 adult patients receiving 100 billion to 1 trillion viruses. And it seems to have worked, depending upon how one assesses success.

?The triglycerides dropped, but after 60 days they trended back up. The primary endpoint had failed, but the secondary endpoint was recurring episodes of pancreatitis ? and they found a statistically significant, or close to it, decrease,? explained James Wilson, MD, PhD, editor-in-chief of Human Gene Therapy and professor of pathology and laboratory medicine at the University of Pennsylvania, who developed the vector. Tracking a few more patients, work not yet published, may have led the EMA?s Committee for Medicinal Products for Human Use to finally recommend approval, after three rejections.

Tomas Salmonson, MD, acting chair of the committee, points to the new data as well as restricting use to the sickest patients in pushing the gene therapy forward. ?Our established ways of assessing the benefits and risks of Glybera were challenged by the extreme rarity of the condition and also by uncertainties associated with data provided.?

For the additional study, the researchers looked at what was happening in the chylomicrons in the blood, and found that triglyceride level can fluctuate, contrary to assumptions of steady change. And that means something is happening that might explain the decrease in the painful episodes ? a very real measurement. Summed up Jean Bennett, MD, PhD, leader of one of the LCA2 clinical trials at Penn, ?It?s a huge vote of confidence for the entire field of gene transfer.?

Dr. Wilson agrees. The repercussions won?t be at the FDA, where scientists make decisions based on data, he said, but on the willingness of big pharma to invest in gene therapy. Despite recent successes ? LCA2, hemophilia, adrenoleukodystrophy ? the pharmaceutical industry has been hesitant to fund gene therapy because it has lacked an approval. ?So-called regulatory uncertainty has been the biggest problem, and if there?s no precedent, they can continue to say no. Biopharma is not interested in the ultra orphans. But I have a feeling we?ll be seeing some activity,? he added.

#2: FINDING HOMES FOR STALLED DRUGS

By August 14, researchers can submit pre-applications to the National Center for Advancing Translational Sciences (NCATS) Discovering New Therapeutic Uses for Existing Molecules program. The idea is simple yet brilliant: match compounds that are languishing on company shelves to diseases with newly-discovered mechanisms. Such candidate drugs have passed initial safety tests but were dropped for business reasons, such as a tiny market, or because they didn?t treat what they were intended to.

Since the announcement in June, eight industry leaders have signed on, offering an initial 58 compounds to find new therapeutic homes. And the need is compelling: of the 4,500+ diseases with recently-revealed mechanisms, only about 250 have treatments. ?If researchers funded through this effort can demonstrate new uses for the compounds, they could significantly reduce the amount of time it takes to get a treatment to patients in need,? said Kathy L. Hudson, PhD, NCATS acting deputy director.

Everyone wins.

#3: SPEEDING FDA APPROVAL

On July 9, President Obama signed into law the FDA Safety and Innovation Act, which updates the 1983 Orphan Drug Act. The new law provides $6 billion over the next 5 years to assist the agency in evaluating new drugs and medical devices. The Act will speed access to new treatments and development of especially promising ones, and the Humanitarian Use Devices program will target those that treat rare diseases, giving priority to diseases of children. ?Treatments are desperately needed because most are serious, many are life-threatening, and about two-thirds of the patients are children,? said Peter L. Saltonstall, president and CEO of the National Organization for Rare Disorders (NORD), which was critical in developing both acts.

The Act may be a lifesaver for people such as 8-year-old Hannah Sames, one of 54 people in the world known to have giant axonal neuropathy. The gene therapy trial that she will take part in is nearing phase 1, but the sponsoring not-for-profit, Hannah?s Hope Fund, is about to run out of money.

#4: EASING INSURANCE ACCESS

When the Supreme Court upheld the Affordable Care Act on June 28, I scrolled through the relieved statements from various rare disease organizations. Thanks to the ACA, children like Hannah Sames and Gavin Stevens will not be penalized for their pre-existing conditions, nor face annual or lifetime insurance caps.

#5: IDENTIFYING DISEASE GENES

Exome sequencing can identify mutations when single-gene tests don?t. The strategy sequences the protein-encoding part of the human genome in individuals, usually young children, whose syndrome has evaded recognition, searching for mutations passed silently from parents, with functions that could explain the symptoms. Once that?s known, researchers can develop new treatments, or repurpose existing ones.

New exome-derived discoveries are being reported nearly weekly, some appearing in the media before the technical papers are published. A recent news release about a 4-year-old named Maya with a neurological disease, for example, made its way into many news reports and blogs, with a touching story and accolades. Yet none named the gene or its precise function ? the part I?m most interested in.

In contrast to the incomplete Maya story, when John Chiang, PhD, director of the Molecular Diagnostics Laboratory at the Casey Eye Institute in Portland, Oregon told me he?d discovered Gavin Stevens? mutation among nearly 2,500 gene variants in the blind boy?s exome, he asked that I not report it. That was 8 months ago ? the mutation is unveiled in a quartet of papers in the current Nature Genetics, after something of a turf war among four research groups.

Gavin?s parents had heard about Dr. Chiang at the Foundation for Retinal Research meeting two years ago, where Jennifer had called him, distraught, after learning that single-gene tests couldn?t explain their son?s blindness. Dr. Chiang, who described his skill as ?I do the dirty work, I find the mutations,? had helped several families after existing tests had fruitlessly, but expensively, probed the most common parts of only the most common genes. Dr. Chiang had first developed larger gene testing panels, and when those still didn?t identify some families? mutations, quietly sent their DNA off to the Beijing Genome Institute for whole exome sequencing.

Now that exome sequencing is commercially available in the U.S., Dr. Chiang cautions that it still doesn?t help all families, and that costs can greatly exceed the oft-mentioned $1,000 pricetag when considering analysis. ?I would only recommend it as the last resort when all known genes are ruled out,? he advised.

On Saturday at the retinal research conference last weekend, I watched Jennifer and Troy beam as Eric Pierce, MD, PhD, director of the Ocular Genomics Institute in Boston and co-author of one of the Nature Genetics papers, talked about their mutation. Discovery of the gene, which affects cellular energy (NAD synthesis), is a starting point for gene therapy, and this particular candidate is a great target. ?The gene is small, and encodes an enzyme,? said Dr. Pierce.

The next day, as my talk about the history of gene therapy wound down, I took stock of my audience. Two young women with canes sat in the front row. A few rows back sat Karen Poulakos, also with a cane, whom I?d chatted with earlier.

Karen has Corey?s disease, LCA2, but, at age 63, had been deemed too old for the gene therapy clinical trial two years ago. But things had changed, she?d learned at the meeting, and she just might be eligible for the phase 3 trial coming up. Karen has lived a full life in her world of shadows, barely remembering when she could see better, and she?s now contemplating what it might be like to see again.

As I collected my things, I marveled at the hope radiating from the faces in the room, sighted as well as not. And I thought that this is science at its very best. This is what it is all about, the molecules, the mice, the deciphering of nature?s mechanisms: helping people.

BANGOR, Maine ? Despite dabbling in filmmaking since the age of 6 and producing and-or directing several short films over the last 10 years, Jacob Caron had never entered a film in a competition.

Knowing about his love and interest for filmmaking, Tam Caron ? one of his aunts ? suggested he produce one and enter it in the 35th annual Maine Student Film and Video Festival.

The only problem was the Brewer High School student had just two weeks to create, edit and submit a film by the festival deadline.

?The good news was the festival rules weren?t too limiting. The bad news was I had two weeks to do it and it was the last two weeks of school,? said Caron, who lives in Eddington. ?But I thought about it and figured I might as well go for it. No guts, no glory.?

Just over a month later, the 17-year-old senior-to-be is basking in the glory after his short film titled ?Freedom of the Press? beat out 29 other entries and won the festival?s grand prize.

?I think there were 29 other film entries. It was kind of overwhelming. When I found out I won, I couldn?t believe it,? Caron said. ?I expected someone else with a more dramatic or thematic film to win.?

Caron?s 13-minute film is part ?Goodfellas,? part ?Untouchables,? part ?Stand By Me? and part ?Doogie Howser, M.D.? without the M.D. It chronicles the activities of the New England Mafia, run by the youngest organized crime boss in the country, and a young, crusading journalist who has been hired by a local newspaper editor to do an expose on the criminal organization.

?This is by far the longest film I?ve done. The previous longest one was seven or eight minutes,? said the son of Drew and Nancy Caron. ?My friends and I put this together in two weeks.?

And they did it under budget. Way under budget.

?My friends acted with payment of a large pizza after the shoot,? Caron said with a laugh. ?I think my total expense was about $30 for clothes at Goodwill.?

Caron not only directed the film, he also acted (playing two parts as a TV reporter and the newspaper editor), filmed, cast and handled most of the production work. He even composed his own musical score.

?All the music for this had to be original. I play piano, so that helped me be able to mix a score for the movie,? he said.

Caron, who also is entering his film in the Portland Film Festival student division, did all the musical scoring on deadline day for the film?s submission.

The film was shot at various locations all over Bangor, Brewer and Orono, including his mother?s optometrist office. It was a marathon effort packed into a tight time frame.

?I could barely keep my eyes open in school some days. We were all exhausted,? said Caron. ?We had several all-nighters, including the last day before the deadline.?

Caron unintentionally bucked a couple of trends with his movie. He went longer than the suggested 10-minute limit and he elected not to go with a drama filled with imagery, enigma or esoteric, outside-the-box themes.

?I wasn?t really too into that, so I figured I?d make something different, and something they hadn?t seen before,? he said.

At the screening of his film last Saturday, it was apparent many in attendance hadn?t seen this before.

?At the screening, there was a big population of children under the age of 10 because they grouped all the kids? films together with the older one,? Caron explained. ?Once the F-bombs started and the lead pipe [used in an implied mob beating scene] came out, the theater doors started opening and kids started leaving.

?There were a few dirty looks from parents and about half the theater cleared.?

Still, Caron, whose older sister Rachel is majoring in performing arts in college, got high marks from the three judges at the competition and won a two-week scholarship to the Maine Media Workshop to learn from filmmakers, editors and videographers from all over the world earlier this month.

?One of the festival officials said he couldn?t believe this kind of film came from a student,? Caron said. ?They liked the cinematography, camera angles and the lighting.?

Not bad for a guy whose school doesn?t even have a film club. That made the workshop an even greater opportunity for someone hoping to major in film study at either the University of Southern California or University of California Los Angeles.

?The whole thing was a really cool experience,? he said. ?Even with all the sleep deprivation.?

OS X Mountain Lion now available in the Mac App Store!

OS X Mountain Lion, the 8th generation of Apple's desktop operating system and the elder brother with which iOS shares much of it's digital DNA, is now available in the Mac App Store.

Most iOS users still use Windows as their primary desktop operating system. Even as the iPad is replacing traditional, lower-level desktop tasks like email, web browsing, and gaming for more and more of the mainstream, when it comes to proper old operating systems, Microsoft is still king.

Yet Apple's choosing an incredibly interesting way to try and change that, at least for the users it cares about most. No, not you, geeky gaming rig guy in the basement. No, not you beleaguered beige box guy in the cubicle. No, not even you, media server maven guy with your house all wired. With Mountain Lion, as with Lion before it, targets the casual user -- that very same mainstream user who's gotten an iPad for many things, but still needs a traditional computer for some things. A car driver looking for a familiarly appointed truck.

Familiarity is the key. Familiarity is a feature. Though they share many of the same underpinnings, OS X and iOS are very different animals. (One's a big cat, for starters.) Yet Apple understands that, for the user, the interface and the experience is the app. So with Mountain Lion, they've made more of the OS X interface and experience more closely match iOS than ever. Which makes sense, given the iOS user base not only dwarfs the OS X user base, but consists of the very users Apple wants to reach most -- those for whom traditional computers have always been inaccessible, intimidating, and off-putting.

Setup and iCloud

OS X Mountain Lion now sets up similarly to iOS 5 -- you enter your iCloud-associated Apple ID and almost everything gets done for you. It's not quite that simple of course, at least not yet, but it's vastly better than computers of even a few years ago.

You still have to choose or take a profile picture, which is irksome to those of us who've used Macs with .Mac and MobileMe for years, and iCloud now, who'd really love to have the option to use our existing iCloud avatar. You also still have to fill out registration information. iOS skips both these steps, and for the better.

Once you're done you're done and in, however, and everything that syncs between your iOS devices also syncs between your iOS devices and Macs. (Apple technically doesn't use sync for iCloud but "store and push".)

All your stuff is "just there", and because Apple has made OS X apps sound, look, and even feel more like iOS apps, all your stuff is easy to find as well.

##Built-in apps

Gone are the iCal and Address Book of old, and in their place are Calendar and Contacts, just like iOS. They got an iPad-style makeover last year with OS X Lion, so this year the name is the biggest change. It may not seem like a big deal, but for an iOS user it makes them instantly recognizable and referable across Apple's platforms.

Likewise, iChat has become Messages... and is more problematic. iMessage delivered a much-needed roshambo to the carrier price gouge that was SMS, and worked in a similar fashion. It was in context. It was understood. Even on the iPad, iMessages made some sense, even if dual-wielders had to put up with dual accounts (phone number for iPhone, Apple ID for iPad and iPod touch) -- and dual alarms! (iOS 6 will fix this by allowing phone numbers and Apple IDs to be merged for iMessage and FaceTime.) On OS X, however, the iMessage/iChat hybrid is messy.

On one hand, if you're working on the Mac, it's faster to respond to iMessages on the Mac than to switch devices and do it on the iPhone or iPad. On the other, holy triple beeping hell, Batman. I've turned off alerts for iMessage on everything but my iPhone, since it's always with me. That way I only get one notification, but can reply from the device of my choosing. A more elegant solution from Apple, however, awaits.

Added to the built in OS X app suite are Notes and Reminders, which now exist and do on OS X what they done on iOS since versions 1 and 5 respectively. They're no more or less than any iOS user would expect, but being able to expect them on the desktop as well as mobile is significant.

The mainstream financial media touts a variety of ?anti-inflation? investments that can be used to protect wealth against the ravages of inflation. These typically range from buying commodities like oil and coal to more sophisticated instruments like inflation-protected treasuries and annuities.

While such investments may appear to be inflation-proof on paper, none of these investment vehicles have the solid intrinsic value of gold, silver or any of the other precious metals.

Intrinsic Value is Everlasting

Intrinsic value does not mean that the product may have value for some time, or even for a long time. Instead, intrinsic value signifies that the item has value forever.

Unlike the steadily declining value of intrinsically worthless paper fiat currency, the worth of an item with true intrinsic value remains relatively unchanged over time and is roughly worth the same amount today as it was thousands of years ago.

Gold and silver are some of the few products with intrinsic value, which would also include necessities of life such as food and shelter. Furthermore, these metals have been in demand for thousands of years for their natural beauty and use in jewelry.
